Darth Vader.

Darth Vader will always be my most evil character, simply because of one reason.

He killed a bunch of children, because some evil dude told him to. I don't know how the hell someone can kill a bunch of kids just because someone tells them to. Darth Vader is also one pretty evil mother dude.

He turns out to be a good guy who just made a bad mistake.

I'm going to have to go with the Joker for this one, though.

The best? I would have to go with the obvious, commonly picked choice, The Joker. However, my favorite villain of all time is Fu Manchu, probably because of my ethnicity but whatever.

There are too many evil characters to choose from. In literature, I'd say Dracula. In movies, the Joker is fantastic. And for real life, I would say...Hitler or Stalin.
