As Killjoy said, right click on the shortcut and select properties > Open File Location. Make sure there's still a firefox.exe there, though I'd doubt there wouldn't be. Try reinstalling Firefox, as this could repair any issues. If you wish to uninstall, make sure that your bookmarks etc are backed up, and if they're not in IE or another browser, use a program like MozBackup to ensure you won't lose your information.
Also, is there a 'firefox.exe' process running in Task Manager?
